RGMA는 신경계 발달 및 성체 신경계에서 다양한 기능을 수행하는 repulsive guidance molecule (RGM) 패밀리의 구성원입니다.
본 단백질은 두부 신경관 폐쇄를 조절하고, 신경돌기 성장과 피질 뉴런 가지 형성을 억제하며, 성숙한 시냅스 형성을 조절합니다.
NEO1/neogenin 수용체와 결합 시 RHOA-ROCK1/Rho-kinase 신호경로를 UNC5B-ARHGEF12/LARG-PTK2/FAK1 연쇄를 통해 활성화하여 신경 성장 원뿔 붕괴 및 신경돌기 성장 억제를 유도합니다.
또한 HRAS-PTK2/FAK1-AKT1 경로에 영향을 미쳐 HRAS 비활성화를 유도합니다.
RGMA는 SMAD1, SMAD5, SMAD8을 통한 BMP 보조수용체로서의 역할도 수행합니다.
