Product Specific Information

The 390 monoclonal antibody reacts with mouse CD31 (PECAM-1, gpIIa), a 130–140 kDa surface protein expressed by endothelial cells and at low levels on leukocytes and platelets. CD31 mediates homotypic interactions important for adhesion, cell-cell and cell-matrix interactions, and signal transduction.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Flow cytometric analysis
• Immunohistochemical staining of frozen tissue sections

Applications Tested:

• Flow cytometric analysis of mouse thymocytes and splenocytes

NovaFluor dye compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer usage:

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample before combining with cells
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures with minimized spillover and spread.

Target Information

CD31 (PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling via ITIM motifs. It is mainly located at junctions between vascular endothelial cells and mediates homophilic binding important for cell-cell interactions. CD31 plays diverse roles in:

• Integrin-mediated cell adhesion
• Transendothelial migration
• Angiogenesis and apoptosis
• Negative regulation of immunoreceptor signaling
• Autoimmunity and macrophage phagocytosis
• IgE-mediated anaphylaxis and thrombosis
