Product Specific Information

The 390 monoclonal antibody reacts with mouse CD31 (PECAM-1, gpIIa), a 130–140 kDa surface protein expressed on endothelial cells, leukocytes, and platelets. CD31 mediates homotypic interactions important for adhesion, cell-cell and cell-matrix interactions, and signal transduction.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications

• Reported for use in flow cytometry and immunohistochemical staining of frozen tissue sections.
• Tested by flow cytometry on mouse thymocytes and splenocytes (≤ 0.6 µg/test).
• Tested by immunohistochemistry on frozen tissue (≤ 20 µg/mL).
• Antibody should be titrated for optimal performance.

Compatibility Notes

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays with NovaFluor dyes.

CellBlox Plus Blocking Buffer

This product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance.

• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor Technology

NovaFluor conjugates are based on Phiton™ technology using engineered nucleic acid dye structures for optimized fluorescent signatures, minimizing spillover and spread.

Target Information

CD31 (PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling through ITIM motifs. It is expressed throughout the vascular compartment, mainly at cell junctions. The N-terminal Ig-like domain mediates homophilic binding crucial for cell-cell interactions. CD31 plays diverse roles in:

• Integrin-mediated adhesion
• Transendothelial migration
• Angiogenesis and apoptosis
• Regulation of immune signaling and thrombosis
