Product Specific Information

The 390 monoclonal antibody reacts with mouse CD31 (PECAM-1, gpIIa), a 130–140 kDa surface protein expressed by endothelial cells and at low levels on leukocytes and platelets. CD31 plays roles in adhesion, cell-cell and cell-matrix interactions, and signal transduction.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Used for flow cytometric analysis and immunohistochemical staining of frozen tissue sections.

Applications Tested:

• Flow cytometry: ≤ 0.6 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
• Immunohistochemistry (frozen tissue): ≤ 20 µg/mL

NovaFluor dyes compatibility:
Not compatible with DNA intercalating dyes (PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample (regardless of NovaFluor count).
For single-color controls: add 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates use Phiton™ technology — engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread.

CD31 (PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling via ITIM motifs. It is expressed throughout the vascular compartment, mainly at cell junctions. The N-terminal Ig-like domain mediates homophilic binding crucial for cell-cell interactions.
Functions include:

• Integrin-mediated adhesion
• Transendothelial migration
• Angiogenesis and apoptosis
• Negative regulation of immune signaling
• Macrophage phagocytosis
• IgE-mediated anaphylaxis and thrombosis
