Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin receptor family. It plays an essential role in signal transduction, T-cell differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, acting as a co-receptor with the T-cell receptor (TCR) for MHC class I molecules. CD8α is critical for MHC-I binding and is also found on NK cells, monocytes, and dendritic cells as homodimers of CD8α. Engagement of MHC-I/peptide complexes leads to recruitment of Lck kinase, inducing cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are vital for pathogen and tumor cell clearance. Differentiation of naïve CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
