Target Information

RPS6KA5 (MSK1) is a serine/threonine protein kinase essential for regulating various cellular processes. It mediates mitogen- or stress-induced phosphorylation of transcription factors such as CREB1 and ATF1 and regulates RELA, STAT3, and ETV1/ER81. It contributes to gene activation via histone phosphorylation and inflammatory gene regulation.
MSK1 phosphorylates CREB1 and ATF1 in response to stimuli such as UV-C irradiation, EGF, and anisomycin. It also controls RELA transcriptional activity in response to TNF and glucocorticoids through cytoplasmic association with NR3C1 (glucocorticoid receptor), repressing inflammatory gene expression.
In skeletal myoblasts, MSK1 is required for RELA phosphorylation at Ser-276 during oxidative stress, and in erythropoietin-stimulated cells, it mediates STAT3 Ser-727 phosphorylation and transcriptional regulation.
