Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j (also known as ILT2, LILRB1, LIR1), a member of the ILT/LIR/MIR receptor family.
CD85j is a transmembrane glycoprotein with four ITIMs that signal through SHP-1 interactions. It is expressed on myeloid cells (monocytes, dendritic cells) and subsets of NK, T, and B cells. Expression correlates with leukemias (ALL, CLL) and viral infections (HIV, EBV, CMV).
Ligands include MHC Class I molecules such as HLA-G, A, F, B27, E, and F.

HP-F1 antibody reduces CD16-dependent cytolytic activity of NK cells.
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on normal human peripheral cells. Recommended ≤5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample prior to combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates are based on Phiton™ technology, utilizing nucleic acid dye structures for engineered fluorescent signatures with minimized spillover and spread.

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosome 19q13.4.
It encodes a receptor with extracellular Ig domains, a transmembrane region, and cytoplasmic ITIM motifs.
Expressed on immune cells, it binds MHC class I molecules, transmitting inhibitory signals that regulate immune activation, inflammation, and cytotoxicity.
Multiple transcript variants and isoforms exist.
