Product Specific Information
The monoclonal antibody HP-F1 recognizes CD85j (ILT2, LILRB1, LIR1), a member of the ILT/LIR/MIR family.
CD85j is a transmembrane glycoprotein with a long cytoplasmic domain containing 4 ITIMs that signal through SHP-1.
Expression is found on myeloid cells (monocytes, dendritic cells) and subsets of NK, T, and B cells.
It is correlated with leukemias (ALL, CLL) and viral infections (HIV, EBV, CMV).
Ligands include MHC Class I molecules such as HLA-G, A, F, B27, E, and F.
HP-F1 has been shown to reduce CD16-dependent cytolytic activity of NK cells.
Each product contains:

- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on human peripheral blood cells
Use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
NovaFluor Compatibility:
- Not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) for optimal staining.
Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor Dyes:
Based on Phiton™ technology utilizing nucleic acid dye structures engineered for minimal spillover and spread.

Target Information
CD85j belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosome region 19q13.4.
It has extracellular immunoglobulin domains, a transmembrane domain, and ITIM motifs that transmit inhibitory signals.
Expressed on immune cells, binding to MHC class I molecules to regulate immune activation and prevent autoreactivity.
Multiple transcript variants encoding different isoforms exist.
