Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j, also known as ILT2, LILRB1, and LIR1. CD85j is a member of the ILT/LIR/MIR family and is a single transmembrane glycoprotein with a long cytoplasmic domain containing four ITIMs that signal through SHP-1 interactions.
Expression is found on myeloid cells (monocytes and dendritic cells) and some lymphoid cells, including subsets of NK, T, and B cells. It has been correlated with leukemias (ALL, CLL) and viral infections such as HIV, EBV, and CMV. Ligands include MHC Class I molecules such as HLA-G, A, F, B27, E, and F.

The HP-F1 antibody reduces CD16-dependent cytolytic activity of NK cells. Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on normal human peripheral cells at ≤5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI). Recommended viability dyes: Invitrogen LIVE/DEAD Fixable Dead Cell stains.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Add 5 µL per sample for optimal staining. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing novel nucleic acid dye structures engineered for minimal spectral spillover and spread.

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at 19q13.4. The encoded protein is part of the subfamily B class of LIR receptors, containing extracellular immunoglobulin domains, a transmembrane domain, and ITIM motifs. It is expressed on immune cells, binds MHC class I molecules, and transduces inhibitory signals to regulate immune responses and prevent autoreactivity. Multiple transcript variants encoding different isoforms exist.
