Product Specific Information

Description:
Monoclonal antibody HP-F1 recognizes CD85j (also known as ILT2, LILRB1, LIR1), a member of the ILT/LIR/MIR receptor family. CD85j is a transmembrane glycoprotein with four ITIMs interacting with SHP-1, expressed on myeloid cells (monocytes, dendritic cells) and subsets of NK, T, and B cells. Expression correlates with leukemias (ALL, CLL) and viral infections (HIV, EBV, CMV). Ligands include MHC Class I molecules (HLA-G, A, F, B27, E, F). HP-F1 reduces CD16-dependent NK cytolytic activity.

Each product includes:

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL sample (10³–10⁸ cells).

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosome 19q13.4. The encoded protein is a B-class LIR receptor with extracellular immunoglobulin domains, a transmembrane region, and cytoplasmic ITIMs. Expressed on immune cells, it binds MHC class I molecules and transduces inhibitory signals to regulate immune activation, inflammation, and cytotoxicity. Multiple isoforms exist due to transcript variants.
