Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j (ILT2, LILRB1, LIR1), a member of the ILT/LIR/MIR family.
CD85j is a single transmembrane glycoprotein with four ITIMs that interact with SHP-1 for inhibitory signaling.
It is expressed on myeloid cells (monocytes, dendritic cells) and some lymphoid cells (NK, T, B cell subsets).
Expression correlates with leukemias such as ALL and CLL and plays a role in viral infections (HIV, EBV, CMV).
Ligands include MHC Class I molecules (HLA-G, A, F, B27, E, F).
HP-F1 antibody reduces CD16-dependent cytolytic activity of NK cells.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for human peripheral blood cells at 5 µL (0.25 µg) per test.
A test is defined as the amount of antibody that will stain a cell sample in 100 µL final volume.
Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample to antibody cocktails before mixing with cells.
    • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence with reduced spillover and spread.

Target Information

The CD85j gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosome 19q13.4.
It encodes a subfamily B class receptor with 2–4 extracellular Ig domains, a transmembrane region, and cytoplasmic ITIMs.
Expressed on immune cells, it binds MHC class I molecules and transduces inhibitory signals to regulate immune activation.
This receptor modulates inflammatory responses and cytotoxicity, helping prevent autoreactivity.
Multiple transcript variants encoding different isoforms have been identified.
