Product Specific Information

The monoclonal antibody L26 recognizes human CD20, a two-transmembrane spanning protein found on precursor and mature B lymphocytes. Expression is lost upon differentiation to plasma cells. CD20 levels are critical for evaluating malignant B lymphocytes including Non-Hodgkin’s Lymphoma and various B-cell leukemias.

The L26 antibody recognizes an epitope in the cytoplasmic domain, requiring tissue permeabilization for staining. Unlike antibodies recognizing extracellular domains, L26 binding is not blocked by therapeutic antibodies such as Rituximab.

Applications Reported:

• Immunohistochemical staining of frozen and formalin-fixed paraffin-embedded tissue sections.

Applications Tested:

• Tested on human FFPE tonsil with low pH antigen retrieval and effective at ≤10 µg/mL.
• Can detect subpopulations of CD20-positive cells without antigen retrieval.
• High pH antigen retrieval possible but may reduce morphology quality.
• Optimized for immunohistochemistry; not recommended for flow cytometry.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 associates with lipid rafts and remains surface-bound after antibody treatment, making it a valuable therapeutic target for B-cell depletion.
Dysfunction of CD20 is linked to Ms4a1-related common variable immune deficiency.
