ERp72는 세 개의 CGHC 활성 부위를 가지며, 이는 단백질 이황화 결합 재배열 과정에 중요한 역할을 합니다.
또한 C-말단에 ER 유지 신호(KEEL)를 포함하고 있으며, 분자 샤페론(molecular chaperone)으로 기능합니다.
