Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding allows ER to regulate gene transcription through coactivator or corepressor recruitment.
There are two forms of the estrogen receptor, alpha (ESR1) and beta (ESR2), encoded by separate genes. At least four estrogen receptor-alpha isoforms are known (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed in various tissues and are essential for sexual development, reproductive function, and bone physiology. They are also implicated in breast cancer, endometrial cancer, and osteoporosis.
