Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational change enabling dimerization and interaction with estrogen response elements (ERE) in DNA. This binding regulates gene transcription by recruiting coactivator or corepressor proteins.
Two forms of the estrogen receptor exist—alpha and beta—encoded by separate genes (ESR1 and ESR2). Alternative RNA splicing produces at least four known estrogen receptor-alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed and crucial for sexual development and reproductive function, as well as roles in bone and other tissues. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
