Product Specific Information

Description:
The 16-10A1 monoclonal antibody reacts with mouse CD80 (B7-1), a 55 kDa member of the Ig superfamily. CD80 is expressed by macrophages, dendritic cells, activated B cells, and activated T cells. It binds to T cell surface antigens CD28 and CD152 (CTLA-4), playing a key role in T-B cell communication and activation.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using stimulated mouse splenocytes. Recommended ≤0.25 µg per test (in 100 µL final volume). Optimal titration is advised.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended detection filter: 660/20 bandpass.

Staining Recommendations:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect from light to avoid photo-induced oxidation.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD80 (B7-1) and CD86 (B7-2) are ligands for T cell costimulatory molecule CD28 and inhibitory receptor CTLA-4 (CD152). Both are expressed on professional antigen-presenting cells and are essential for T cell activation. CD80 is rapidly induced on activated B cells and expressed in various lymphoma cell lines and monocytes. Dysregulation of CD80 is associated with gallbladder squamous cell carcinoma and myocarditis.
