Product Specific Information

Description:
The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor of the B7 family. CD86 is expressed on B cells, macrophages, and dendritic cells, and is upregulated by stimuli such as BCR complex, CD40, and cytokine receptors. It acts as a counter-receptor for CD28 and CTLA-4, playing a critical role in T-B cell crosstalk and T cell costimulation.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test in 100 µL final volume. Cell count per test: 10⁵–10⁸ cells (empirically determined).

Fluorochrome Information:
Super Bright 645 is a tandem dye excited at 405 nm (violet laser) and emits at 645 nm. Recommended detection filter: 660/20 bandpass. Ensure the instrument supports this fluorochrome.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect the vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency.

Target Information

CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation. Insufficient co-stimulation via CD86/CD80 can induce tolerance. CD86 is a type I membrane protein in the immunoglobulin superfamily, expressed by antigen-presenting cells. It binds CD28 (activating T-cells) and CTLA-4 (inhibiting T-cell activation). Alternative splicing produces multiple isoforms. CD86 dysfunction is associated with diseases such as gallbladder squamous cell carcinoma and myocarditis.
