Product Specific Information

The OV9A2 monoclonal antibody reacts with human CD59 (Protectin), a GPI-linked membrane glycoprotein homologous to the murine Ly-6 superfamily. CD59 is expressed on all human lymphocytes, monocytes, granulocytes, erythrocytes, and non-hematopoietic cells such as endothelial cells and neurons.

CD59 binds complement components C8 and C9, inhibiting membrane attack complex assembly and complement-mediated cytolysis. It also interacts with CD2 to modulate T cell adhesion and activation. Altered CD55/CD59 expression is observed in systemic lupus erythematosus (SLE) patients.

Crossblocking studies show OV9A2 binds the same epitope as MEM-43.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by a 405 nm violet laser and emits at 645 nm. Recommended filter: 660/20 bandpass.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to reduce non-specific polymer interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer ([Product # 00-8222-49]) or 1-step Fix/Lyse Solution ([Product # 00-5333-57]) with minimal impact on brightness and FRET efficiency.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Target Information

CD59 (Protectin) is a small (18–20 kDa) GPI-anchored inhibitor of the membrane attack complex (MAC). It prevents autologous cell damage by blocking complement cascade terminal steps and inhibiting C9 incorporation into the complex. CD59 also acts as a low-affinity CD2 ligand, contributing to T cell co-stimulation and signal transduction.
Mutations in CD59 cause CD59 deficiency, associated with hemolytic anemia, thrombosis, and cerebral infarction. Multiple alternatively spliced transcript variants exist.
