Preparation:
Produced from sera of goats immunized with highly pure Recombinant Human sFas Ligand. Anti-Human sFas Ligand-specific antibody was purified by affinity chromatography employing an immobilized Human sFas Ligand matrix.

Sandwich ELISA:
To detect Human sFas Ligand by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. In conjunction with PeproTech Biotinylated Anti-Human sFas Ligand (500-P184GBt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human sFas Ligand.

Western Blot:
For detection of Human sFas Ligand by Western Blot, use at 0.1–0.2 µg/mL. With compatible secondary reagents, detection limit for Recombinant Human sFas Ligand is 1.5–3.0 ng/lane under both reducing and non-reducing conditions.

Target Information

CD178 (Fas ligand, FasL) is a type-II membrane protein with its N-terminus in the cytoplasm and C-terminal region extending extracellularly. Its receptor, FasR, belongs to the TNF and NGF receptor family. CD178 induces apoptosis upon interaction with FasR and exists as membrane-bound (45 kD) or soluble (26 kD) forms, the latter released by metalloproteinase cleavage.

Binding of CD178 to Fas triggers receptor oligomerization and apoptotic cell death. CD178 is mainly expressed in activated T-lymphocytes and NK cells, as well as immune-privileged tissues (testis, eye). Expression also occurs in thymus, liver, ovary, lung, heart, and kidney. CD178-mediated apoptosis is involved in antiviral immune responses and immune regulation, with implications in diseases such as AIDS, hepatitis, cancer, atherosclerosis, and inflammatory disorders.
