Preparation:
Produced from sera of goats immunized with highly pure Recombinant Human sFas Ligand. Anti-Human sFas Ligand-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect hFasL/Apo1L by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required.
This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human FasL/Apo1L (500-P184G) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant hFasL/Apo1L.

Western Blot:
To detect Human sFas Ligand by Western blot analysis, this antibody can be used at 0.1–0.2 mg/mL.
Used with compatible secondary reagents, the detection limit for Recombinant Human sFas Ligand is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

CD178 (Fas ligand, FasL) is a type-II membrane protein with its N-terminus in the cytoplasm and C-terminal region extending extracellularly. Its receptor, FasR, is a cell-surface type-I membrane protein and a member of the TNF and NGF receptor family.
As a member of the TNF-cytokine family, CD178 induces apoptosis upon interaction with FasR. CD178 exists in membrane-bound (45 kD) or soluble (26 kD) forms, with the soluble form released via metalloproteinase cleavage.
Binding of CD178 to Fas triggers receptor oligomerization and apoptotic signaling. It is predominantly expressed in activated T-lymphocytes and NK cells, as well as in immune-privileged tissues (testis, eye).
Expression has also been reported in thymus, liver, ovary, lung, heart, and kidney. CD178-mediated apoptosis plays a key role in antiviral immune responses.
The Fas/CD178 system is implicated in diseases such as AIDS, hepatitis, cancer, atherosclerosis, and inflammatory disorders.
