The monoclonal antibody mhalx21 reacts with mouse interleukin-21 (IL-21), a 17 kDa immunomodulatory cytokine produced mainly by NKT, Th17, and Tfh cells.
IL-21 signaling through IL-21R and STAT3 induces transcriptional activation by Bcl6, critical for Tfh cell effector function.
In Th17 cells, IL-21 promotes autocrine feedback via STAT3, IRF4, and RORγt, upregulating IL-23R and downregulating Foxp3.
High IL-21 levels are found in colitis models; IL-21-deficient mice show protection against chemically induced colitis.
Clone mhalx21 recognizes a different epitope than anti-mouse IL-21 clone FFA21 (Product #51-7211).

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on Th17-polarized mouse splenocytes using IC Fixation and Permeabilization Buffers (Product #88-8823-88).
Use ≤ 0.5 µg per test (100 µL final volume). Optimize by titration for best results.

Target Information

IL-21 is a 17 kDa cytokine produced by NKT, Th17, and Tfh cells. It mediates autocrine signaling via IL-21R and STAT3, activating Bcl6 and supporting Tfh development.
IL-21 regulates B cell differentiation, IgG production, and apoptosis in naive B cells. In Th17 cells, IL-21 promotes IL-23R expression and downregulates Foxp3.
Elevated IL-21 levels are observed in chemically induced colitis; IL-21 deficiency confers protection.
