Product Specific Information

This FFA21 monoclonal antibody reacts with mouse interleukin-21 (IL-21), a 17 kDa immunomodulatory cytokine produced mainly by NKT, T helper (Th) 17, and T follicular helper (TFH) cells.
In TFH cells, IL-21 expression leads to autocrine signaling through IL-21R and STAT3, activating Bcl6 transcription. Similar to IFN-γ for Th1 and IL-4 for Th2, IL-21 is critical for TFH cell effector function. It promotes B cell differentiation into plasma and memory cells, stimulates IgG production, and induces apoptosis in naive B cells.

In Th17 cells, IL-21 expression and feedback via STAT3, IRF4, and RORγt upregulate IL-23R, promoting Th17 maturation by IL-23. IL-21 also downregulates Foxp3. Elevated IL-21 levels are observed in chemically induced colitis models, while IL-21-deficient mice show resistance due to impaired Th17-associated molecule expression.

Preliminary data suggest clone FFA21 recognizes a different epitope than anti-mouse IL-21 clone mhalx21 (Product # 51-7213-82).

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by intracellular staining and flow cytometry of Th17-stimulated mouse cells (≤0.25 µg/test; 100 µL final volume; 10⁵–10⁸ cells/test). Optimize titration for best performance.

Target Information

IL-21 is a 17 kDa cytokine produced mainly by NKT, Th17, and TFH cells. In TFH cells, IL-21 induces autocrine signaling via IL-21R and STAT3, promoting Bcl6 activation. IL-21 is essential for TFH cell development and B cell differentiation into plasma and memory cells, IgG production, and apoptosis induction in naive B cells.
In Th17 cells, IL-21 upregulates IL-23R through STAT3, IRF4, and RORγt, and downregulates Foxp3. IL-21-deficient mice are resistant to colitis induction, while elevated IL-21 is observed in colitis models.
