The sirx6 monoclonal antibody reacts with mouse CD196 (CCR6).
CD196 is a seven-transmembrane G protein-coupled receptor expressed on T, B, dendritic, NK, and Langerhans cells. It binds MIP3a/CCL20, a chemoattractant for dendritic cells, effector/memory T cells, and B cells. CD196 is involved in host defense and inflammation at epithelial sites and plays a role in Th17 differentiation and CD4⁺FoxP3⁺ regulatory T cell development.

Due to possible receptor shedding or intracellular cycling, fixation after surface staining is recommended.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes. Recommended ≤1 µg/test.
Cells should be surface stained, washed, fixed with IC Fixation Buffer for 20–30 min, washed again, and analyzed.
A test is defined as staining a 100 µL sample, typically containing 10⁵–10⁸ cells/test.
Optimize antibody titration for best performance.

Target Information

CCR6 belongs to the beta chemokine receptor family and encodes a seven-transmembrane G protein-coupled receptor. It is preferentially expressed by immature dendritic cells and memory T cells. The ligand, MIP-3α (CCL20), mediates B-lineage maturation and antigen-driven B-cell differentiation. CCR6 regulates migration and recruitment of dendritic and T cells during immune and inflammatory responses. Alternative splicing variants encoding the same protein have been described.
