Product Specific Information
The eBio64DEC17 antibody reacts with human IL-17A and functions as a neutralizing antibody. IL-17A is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
Human IL-17A exists as glycosylated 20–30 kDa homodimers produced by activated CD4+ T-cells. It enhances ICAM-1 expression in fibroblasts and stimulates secretion of IL-6, IL-8, G-CSF, and PGE2 from epithelial and endothelial cells. Fibroblasts exposed to IL-17A support CD34+ progenitor proliferation and neutrophil maturation.
IL-23-dependent Th17 cells produce IL-17A, distinct from Th1/Th2 pathways. IFNγ and IL-4 negatively regulate Th17 cell generation. Intracellular staining by eBio64DEC17 identifies the same population as eBio64CAP17.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining and flow cytometry of stimulated human peripheral blood cells. Use at 5 µL (0.03 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

eFluor® 660 replaces Alexa Fluor® 647. Emission at 659 nm, excitation with red laser (633 nm).

Target Information
Interleukin-17A (IL-17A, CTLA-8) is a 32 kDa disulfide-linked homodimer (136 amino acids) that signals through IL-17R/CDw217. It regulates NF-κB and MAPK pathways, enhancing IL-6 and COX-2 expression and nitric oxide production. IL-17A family includes IL-17A–F proteins. High IL-17A levels are linked to inflammatory and autoimmune conditions.
