Product Specific Information

The BM8 monoclonal antibody reacts with mouse F4/80 antigen, an approximately 160 kDa surface receptor belonging to the EGF-TM7 family. It contains seven EGF-like domains, seven transmembrane sequences, and a cytoplasmic domain homologous to TM7 proteins. The F4/80 antigen is a key marker for mature macrophages but is also found on peritoneal eosinophils, Langerhans cells, and certain dendritic cell subsets. Expression begins early in myeloid development and can be upregulated by M-CSF stimulation. Some macrophage populations, particularly in lymphoid tissues, may lack F4/80 expression.

Applications Reported:
This antibody is suitable for immunohistochemistry (frozen and paraffin-embedded tissues) and microscopy.

Applications Tested:
Validated for IHC with formalin-fixed paraffin-embedded tissue using pronase antigen retrieval at ≤5 µg/mL. Optimal titration is recommended.

Filter Recommendation:
Use a filter capturing the 570 nm emission wavelength (e.g., Excitation 545/25, 565LP, Emission 605/70). Compatible with standard Alexa Fluor® 555 or TRITC filters.

Target Information

F4/80 antigen is a 160 kDa cell surface glycoprotein, part of the EGF-TM7 family, sharing 68% amino acid identity with human EMR1. Expression varies with macrophage maturation and activation, found in Kupffer cells, Langerhans cells, microglia, gut macrophages, and others. Absent in macrophages of T-cell areas in spleen and lymph nodes. The biological function remains under study, but evidence suggests a role in generating efferent CD8⁺ regulatory T cells.
