Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human TNF-alpha. Anti-Human TNF-alpha-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect Human TNF-alpha by sandwich ELISA (using 100 µL/well antibody solution), use 0.25–1.0 µg/mL of this antibody. In conjunction with PeproTech Polyclonal Anti-Human TNF-alpha (500-P31A) as a capture antibody, it enables detection of at least 0.2–0.4 ng/well of Recombinant Human TNF-alpha.

Western Blot:
To detect Human TNF-alpha by Western Blot, use 0.1–0.2 µg/mL. Detection limit for Recombinant Human TNF-alpha is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers but shows a single 17 kDa band in SDS-PAGE under non-reducing conditions. TNF alpha is involved in immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor necrosis. It has implications in autoimmune diseases, insulin resistance, and cancer.
