Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human TNF-alpha. The antibody was purified by affinity chromatography using an immobilized Human TNF-alpha matrix.
Sandwich ELISA:
For detection of Human TNF-alpha by sandwich ELISA (100 µL/well antibody solution), use 0.5–2.0 µg/mL. When used with PeproTech Biotinylated Anti-Human TNF-alpha (500-P31ABt) as detection antibody, it can detect 0.2–0.4 ng/well of Recombinant Human TNF-alpha.
Western Blot:
For detection of Human TNF-alpha, use at 0.1–0.2 µg/mL. Detection limit: 1.5–3.0 ng/lane under reducing or non-reducing conditions.
Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers but appears as a single 17 kDa band in SDS-PAGE under non-reducing conditions. TNF alpha exhibits tumor necrosis activity, plays roles in immune responses to infections, and is implicated in autoimmune diseases, insulin resistance, and cancer.
