Product Specific Information
Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems developed by cloning specific antibody DNA sequences from immunoreactive rabbits. Individual clones are screened to select optimal candidates for production.
Advantages include:
- High specificity and sensitivity
- Lot-to-lot consistency
- Animal origin-free formulation
- Broad immunoreactivity due to the large rabbit immune repertoire
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, which are involved in G1 or G2/M arrest.
The p53 structure includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerization domain
- C-terminal regulatory domain
Phosphorylation at Ser15 and Ser392 plays critical roles in p53 activation and stabilization. Ser392 phosphorylation is associated with tumor formation. p53 also influences aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP).
Mutations in p53, common in various cancers, impair DNA binding and tumor suppressor function. TP53 gene alterations occur both as somatic and germline mutations, including in Li-Fraumeni syndrome.
