세포 내 위치 (Cellular Localization)
Cytoplasm and nucleus. In the absence of TGF-β, localized in both cytoplasm and nucleus. Upon TGF-β stimulation, migrates to the nucleus when complexed with SMAD4. Dephosphorylation by PPM1A releases it from the SMAD2/SMAD4 complex and exports it from the nucleus via RANBP1 interaction. During early embryonic stages, mainly localized to the nucleus, later becoming evident in the cytoplasm at blastocyst and epiblast stages.
배경 (Background)
The encoded protein belongs to the SMAD family, homologous to Drosophila mothers against decapentaplegic (Mad) and C. elegans Sma genes. SMAD proteins act as signal transducers and transcriptional modulators mediating multiple signaling pathways. This protein mediates TGF-β signaling, regulating cell proliferation, apoptosis, and differentiation. It interacts with SARA protein to be recruited to TGF-β receptors, where it becomes phosphorylated. Phosphorylation induces dissociation from SARA and association with SMAD4, facilitating nuclear translocation and transcriptional regulation.
