Product Specific Information

This antibody detects endogenous levels of Tau only when phosphorylated at Thr498, which site has been historically referenced as Thr181.

Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting Tau acts as a linker between these structures.

Axonal polarity is influenced by Tau/MAPT localization, with short isoforms contributing to cytoskeletal plasticity and longer isoforms aiding stabilization. In its hyper-phosphorylated form, Tau is the major component of paired helical filaments (PHF), which are central to neurofibrillary lesions in Alzheimer’s disease (AD). Hyper-phosphorylation disrupts Tau’s microtubule binding, leading to neuronal degeneration.

Tau phosphorylation involves multiple kinases, including GSK-3β, PKA, cdk5, and casein kinase II. Hyper-phosphorylated Tau is also found in other neurodegenerative disorders such as Pick’s disease, frontotemporal dementia, cortico-basal degeneration, and progressive supranuclear palsy.
