Description:
This GzA-3G8.5 monoclonal antibody reacts with mouse Granzyme A, the most abundantly expressed of the ten granzyme serine proteases identified in mice. Granzymes are proteins released from NK cells and cytotoxic T lymphocytes that induce death in target cells by cleavage of intracellular substrates. They play a critical role in immune defense against viruses, tumors, and intracellular bacteria.

Granzyme A activates caspase-independent cell death pathways similar to apoptosis, characterized by mitochondrial and DNA damage. It may also play a role in inflammation, as the precursor form of IL-1β (pro-IL-1β) is among its target substrates. Granzyme A shares overlapping substrate specificity with Granzyme K, which may explain the minimal decrease in cytotoxicity observed in Granzyme A-deficient CTLs.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by intracellular staining followed by flow cytometric analysis of normal mouse splenocytes. Recommended ≤0.125 µg per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody should be titrated for optimal performance.

Target Information

Cytolytic T lymphocytes (CTL) and natural killer (NK) cells recognize and lyse specific target cells bearing nonself antigens, typically resulting from intracellular infection. The protein described here is a T cell- and NK cell-specific serine protease that functions as a key component in the lysis of target cells by CTLs and NK cells.
