Description:
The eBio4B10 monoclonal antibody reacts with mouse and human T-bet. T-bet is a Th1-specific T-box transcription factor critical to the development of the Th1 CD4+ T cell lineage. T-Bet deficient mice show impaired Th1 cell development, while ectopic expression of T-Bet skews development toward the Th1 lineage. T-Bet expression is induced by IFN-γ and regulates IFN-γ, IL-12Rβ, and IL-2 expression, influencing DNA accessibility and histone remodeling. It also contributes to class-switch recombination in B cells.

Applications Reported:
This antibody is reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells using the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00).
Recommended use ≤ 0.25 µg per test (100 µL final volume). Cell count may range from 10⁵–10⁸ cells/test. Optimal titration recommended.

Target Information

T-bet (TBX21) is a T-cell specific transcription factor with a single T-box DNA-binding domain. It regulates the expression of the Th1 cytokine interferon-gamma and plays a key role in Th1/Th17 differentiation and immune regulation. T-bet cooperates with Stat4 in programming Th1 gene expression. Genetic variations in T-bet are linked to aspirin-induced asthma and nasal polyps.
