The SH2.1 monoclonal antibody reacts with mouse Siglec-G (sialic acid-binding immunoglobulin-like lectin-G).
Siglec-G is an inhibitory receptor expressed in B cells, with B1 B cells showing the highest expression levels. Overexpression inhibits B cell receptor-mediated calcium signaling. Siglec-G-deficient mice exhibit expanded B1a cell populations and elevated natural IgM titers. These cells show reduced apoptosis and prolonged lifespan, associated with higher NFATc1 expression.
Negative regulation by Siglec-G contributes to the muted signaling response typical of B1 cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Siglec-G functions as an inhibitory receptor in B cells, particularly B1 B cells. Overexpression suppresses B cell receptor-mediated calcium signaling. Siglec-G deficiency leads to increased B1a cells, higher IgM titers, reduced apoptosis, and extended cell lifespan, likely due to enhanced NFATc1 expression. This receptor mediates negative regulation in B1 cells, explaining their subdued signaling behavior.
