Target Information
ICAD (Inhibitor of Caspase-Activated DNase) is a substrate controlling nuclear apoptosis. It exists in two isoforms:
- ICAD-L/DFF45 (45kDa): Functional isoform
- ICAD-S/DFF35 (35kDa): Non-functional isoform
ICAD inhibits CAD (Caspase-Activated DNase) and acts as its folding chaperone. During apoptosis, ICAD is cleaved by caspase-3 at residues 117 and 224, releasing CAD to execute DNA fragmentation. ICAD has been identified as a potential therapeutic target for restoring normal apoptotic signaling in colon and brain cancer cells. Two alternatively spliced transcript variants encode distinct isoforms of the ICAD gene.
