Target Information

ICAD (Inhibitor of Caspase-3-Activated DNase) is a substrate controlling nuclear apoptosis. It exists in two isoforms:

• ICAD-L/DFF45 (45 kDa) – functional isoform
• ICAD-S/DFF35 (35 kDa) – shorter isoform

Both isoforms can bind and inhibit CAD, but only ICAD-L is functional. ICAD is cleaved and inactivated to allow caspase-activated DNase (CAD) to mediate nuclear apoptotic DNA fragmentation. In non-apoptotic cells, CAD is complexed with ICAD, and activation occurs through caspase-3-mediated cleavage at residues 117 and 224, releasing ICAD fragments from CAD.

ICAD also acts as a CAD-specific folding chaperone. Recent studies suggest ICAD as a potential target for restoring apoptotic signaling in colon and brain cancer cells. Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
