Target Information

ICAD is the inhibitor of caspase-3-activated DNase, a substrate that controls nuclear apoptosis. ICAD has two isoforms: a functional isoform of 45 kDa (ICAD-L/DFF45) and a 35 kDa isoform (ICAD-S/DFF35). Both isoforms can bind and inhibit CAD, but only ICAD-L is functional. ICAD is cleaved and inactivated to allow caspase-activated DNase (CAD) to execute nuclear apoptotic DNA fragmentation. In non-apoptotic cells, CAD is complexed with ICAD. Activation occurs through caspase-3-mediated cleavage at residues 117 and 224, releasing ICAD fragments from CAD. ICAD also acts as a CAD-specific folding chaperone. Recent studies suggest ICAD as a potential target for restoring apoptotic signal transduction in colon and brain cancer cells. Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
