Target Information
ICAD (Inhibitor of Caspase-3-Activated DNase) controls nuclear apoptosis by inhibiting CAD (Caspase-Activated DNase).
ICAD has two isoforms:
- ICAD-L/DFF45 (45 kDa): Functional isoform
- ICAD-S/DFF35 (35 kDa): Non-functional isoform
ICAD inhibits CAD until caspase-3 cleaves ICAD at residues 117 and 224, releasing CAD to execute apoptotic DNA fragmentation.
Beyond DNase inhibition, ICAD acts as a CAD-specific folding chaperone.
Recent studies suggest ICAD may serve as a therapeutic target for restoring normal apoptotic signaling in colon and brain cancer cells.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
