Target Information

TLR5 is a Type I membrane protein belonging to the Toll-like receptor family and participates in the innate immune response to microbial agents. It mediates detection of bacterial flagellins and acts via MyD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and the inflammatory response.
TLR5 is highly expressed in liver, detected in lung, and at very low levels in most other tissues. The TLR5 gene locus is associated with susceptibility to Salmonella.
Inbred mouse strains show varying resistance to S. typhimurium:

  • Susceptible: BALB/c, C57BL/6, C3H/He
  • Intermediate: DBA/2, C75L
  • Resistant: A, CBA
    The strain MOLF/Ei is highly susceptible, has a unique TLR5 haplotype, and lower expression of TLR5.
    A common TLR5 stop codon polymorphism disrupting TLR5 signaling is linked to susceptibility to Legionnaires’ disease, highlighting TLR5’s role in innate immunity.
