Target Information
TLR5 is a Type I membrane protein belonging to the Toll-like receptor family, participating in the innate immune response to microbial agents. It mediates the detection of bacterial flagellins and acts via MyD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and inflammatory response.
TLR5 binds to TIRAP and MyD88 via their respective TIR domains. It is highly expressed in liver, detected in lung, and present at low levels in most other tissues.
The TLR5 gene locus is associated with susceptibility to Salmonella. Mouse strains differ in resistance to S. typhimurium infection:
- Susceptible: BALB/c, C57BL/6, C3H/He
- Intermediate: DBA/2, C75L
- Resistant: A, CBA
The MOLF/Ei strain is highly susceptible, has a unique TLR5 haplotype, and lower expression of TLR5.
A common TLR5 stop codon polymorphism disrupting TLR5 signaling is linked to susceptibility to Legionnaires’ disease, highlighting its role in innate immunity.
