Product Specific Information

The Ber-H2 monoclonal antibody reacts with human CD30, a 120 kDa transmembrane glycoprotein of the TNFR superfamily. CD30 is expressed on normal activated T and B lymphocytes, but not on resting lymphocytes. It is also found on certain lymphomas, embryonal carcinomas, and virus-infected lymphocytes (HIV, HTLV, EBV). Interaction with its ligand CD30L leads to signaling and activation of NF-kappaB, regulating apoptosis.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.06 µg) per test (final volume 100 µL). Cell number range: 10⁵–10⁸ cells/test.

Excitation: 488–561 nm
Emission: 578 nm
Laser Compatibility: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered

Target Information

CD30 (Ki-1, TNFRSF8) is a type I transmembrane glycoprotein of the TNF receptor superfamily. Originally identified on Hodgkin’s and Reed-Sternberg cells, CD30 binds CD30L (CD153) to mediate effects including proliferation, activation, differentiation, and apoptosis. It plays a key role in Hodgkin’s disease and CD30+ lymphomas, acting as a costimulatory molecule in thymic negative selection.

CD30 expression is found in:

  • Hodgkin’s and Reed-Sternberg cells
  • Anaplastic Large Cell Lymphomas (ALCL)
  • Some non-Hodgkin’s lymphomas (e.g., Burkitt’s)
  • Virus-infected T/B cells
  • Activated normal T/B cells

Soluble CD30 (sCD30) serves as a Th2 immune response marker. TRAF2 and TRAF5 interact with CD30, activating NF-kappaB signaling. CD30 positively regulates apoptosis and limits autoreactive CD8 effector T cell proliferation.

Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
