Product Specific Information

The 390 monoclonal antibody reacts with mouse CD31 (platelet-endothelial cell adhesion molecule-1, PECAM-1, gpIIa). This 130–140 kDa surface protein is expressed by endothelial cells and at low levels on leukocytes and platelets. CD31 mediates homotypic interactions critical for adhesion, cell-cell and cell-matrix interactions, and signal transduction.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Flow cytometric analysis and immunohistochemical staining of frozen tissue sections.

Applications Tested:

• Flow cytometry: ≤ 0.6 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
• Immunohistochemistry (frozen): ≤ 20 µg/mL
  Careful titration is recommended for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD31 (PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling through ITIM motifs that recruit protein-tyrosine phosphatases upon phosphorylation. It is widely expressed in the vascular system, mainly at endothelial junctions. The N-terminal Ig-like domain mediates homophilic binding, facilitating cell-cell interactions. CD31 plays roles in:

• Integrin-mediated adhesion
• Transendothelial migration
• Angiogenesis and apoptosis
• Negative regulation of immunoreceptor signaling
• Autoimmunity, macrophage phagocytosis, and thrombosis
