Product Specific Information

The 390 monoclonal antibody reacts with mouse CD31 (PECAM-1, gpIIa), a 130–140 kDa surface protein expressed by endothelial cells and at low levels on leukocytes and platelets. CD31 mediates homotypic adhesion and is involved in cell-cell and cell-matrix interactions, as well as signal transduction.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Flow cytometric analysis
• Immunohistochemical staining of frozen tissue sections

Applications Tested:

• Flow cytometry of mouse thymocytes and splenocytes (≤ 0.6 µg/test, 100 µL total volume)
• Immunohistochemistry of frozen tissue (≤ 20 µg/mL)

Careful titration is recommended for optimal performance.

NovaFluor Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample (10³–10⁸ cells/test)
• Use in antibody cocktails/master mixes before combining with cells

NovaFluor conjugates use Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.
Excitation: 552 nm; Emission: 612 nm; Laser: 561 nm (Yellow).

Strain-specific observation:
Non-specific staining of B cells observed in SJL mice; not in BALB/c or C57BL/6 strains.

Target Information

CD31 (PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling through ITIM motifs. It is widely expressed in the vascular compartment, particularly at cell junctions. The N-terminal Ig-like domain mediates homophilic binding, critical for cell-cell interaction. CD31 plays diverse roles in:

• Integrin-mediated adhesion
• Transendothelial migration
• Angiogenesis and apoptosis
• Negative regulation of immunoreceptor signaling
• Macrophage phagocytosis and thrombosis
