Description:
The JES3-9D7 monoclonal antibody reacts with human interleukin-10 (IL-10).

Applications Reported:
Reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Refer to BestProtocols®: Protocol A for intracellular proteins. Use 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days in dark at 4°C with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488–561 nm / 775 nm; Blue, Green, Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes, with some lymphocyte production. It regulates immune responses and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It enhances B cell survival, proliferation, and antibody production, blocks NF-kappa B activity, and regulates the JAK-STAT pathway. Knockout studies suggest IL-10 is critical for intestinal immune regulation. Mutations are associated with increased susceptibility to HIV-1 infection and rheumatoid arthritis. Related diseases include Graft-Versus-Host Disease and HIV-1.
