Product Specific Information

This NJTEN3 monoclonal antibody reacts with the pro-form of mouse IL-1 beta, a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. Synthesized as a 31 kDa inactive pro-form, it is cleaved into the active 17 kDa protein upon inflammasome activation. IL-1 beta signals via IL-1RI, shared with IL-1 alpha, and plays a key role in innate host defense and acute-phase responses. Its activity can be moderated by IL-1 Receptor Antagonist (IL-1RA).

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Validated on stimulated mouse thioglycolate-elicited peritoneal exudate cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and BestProtocols® Protocol A.
Recommended usage ≤ 0.25 µg/test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test (empirical optimization suggested).

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days in the dark at 4°C.
Excitation/Emission: 488–561 nm / 775 nm (Blue, Green, Yellow-Green Lasers).
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

Interleukin-1 beta (IL-1 beta) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. Synthesized as a 31 kDa inactive pro-form, it requires inflammasome activation and caspase-1 processing for cleavage into the active 17 kDa form. It signals through IL-1RI and IL-1RII, shared with IL-1 alpha, and regulates inflammatory and acute-phase responses. IL-1RA competitively inhibits receptor binding, modulating cytokine activity. Elevated IL-1 beta levels are associated with chronic inflammatory conditions, offering potential therapeutic targets.
