Product Specific Information

The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kD cytokine, and functions as a neutralizing antibody. Mouse IL-4 (also known as BCDF, BCGF-1, BSF-1) is secreted primarily by activated Th2 and natural killer T cells and promotes B cell proliferation and differentiation. In conjunction with CD40 and other signals, IL-4 induces B cell isotype switching and upregulates surface MHC class II and CD23.

Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Validated for intracellular staining and flow cytometric analysis of stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Recommended use ≤ 0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration is advised for optimal performance.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.

Excitation / Emission / Laser:
488–561 nm / 775 nm / Blue, Green, Yellow-Green Laser.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

Interleukin-4 (IL-4) is a pleiotropic cytokine produced by activated T cells, acting as a ligand for the IL-4 receptor. It shares receptor binding with IL-13, contributing to overlapping functions. STAT6 mediates IL-4 signaling. The IL-4 gene is part of a cytokine cluster on chromosome 5q with IL-3, IL-5, IL-13, and CSF2, coordinated by long-range regulatory elements. IL-4 delta 2 is a known splice variant.
