Product Specific Information

The C17.8 antibody reacts with the p40 subunit of IL-12 and IL-23 and neutralizes their bioactivity.
For specific neutralization of IL-23 only, see clone G23-8 (anti-mouse IL-23 p19).

IL-12 is a 70 kDa heterodimeric cytokine composed of p40 and p35 subunits, secreted by activated monocytes, macrophages, and dendritic cells.
IL-23 is a heterodimer composed of p19 and p40 proteins, expressed by activated dendritic cells.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Tested using stimulated mouse thioglycolate-elicited peritoneal exudate cells with the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
• Use ≤ 0.25 µg per test (100 µL final volume).
• Recommended titration for optimal performance.

Light Sensitivity:

• Tandem dye is sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:

• Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.
• Clone-specific performance should be determined empirically.

Excitation / Emission / Laser:

• Excitation: 488–561 nm
• Emission: 775 nm
• Compatible Lasers: Blue, Green, Yellow-Green

Filtration:

• 0.2 µm post-manufacturing filtered.

Target Information

IL-12 is produced by dendritic cells, neutrophils, macrophages, and lymphoblastoid cells upon antigenic stimulation.
It is encoded by IL-12A (p35) and IL-12B (p40) genes.
IL-23 shares the IL-12B subunit and includes IL-23A (p19).

Functional Roles:

• IL-12: Enhances endothelial adhesion molecule-1 expression, directs leukocytes to tumors, facilitates necrosis of tumor tissue.
• IL-23: Stimulates memory T-cells, supports Th17 cell induction and maintenance, promotes pro-inflammatory cytokine production.
