Product Specific Information

Description:
The eBio13A antibody reacts with mouse IL-13, a cytokine mainly produced by Th2 cells and antigen-primed CD8 T cells. IL-13 is involved in allergic inflammation, parasitic clearance, and cancer models, showing either inhibitory or stimulatory activity depending on tumor type. In humans, IL-13 contributes to isotype switching in B cells and down-modulates macrophage activity by reducing pro-inflammatory cytokines (IL-1, IL-6, IL-8, IL-10, IL-12).

Applications Reported:
This antibody is reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining of stimulated mouse splenocytes. Recommended usage ≤ 0.5 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark, with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488–561 nm excitation, 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells. It regulates inflammatory and immune responses, down-regulates macrophage activity, and inhibits production of pro-inflammatory cytokines and chemokines. IL-13 is involved in allergen-induced asthma independent of IgE and eosinophils, regulates B cell maturation and differentiation, up-regulates CD23 and MHC class II, and promotes IgE isotype switching. Structurally, IL-13 resembles IL-3, IL-4, IL-5, and GM-CSF. The IL-13 gene is located on chromosome 5q within a cytokine gene cluster near IL4.
