Target Information
Cyclin D1 (PRAD-1, bcl-1)는 세포 주기 조절에 핵심적인 단백질로, cdk4 및 cdk6과 결합하여 Rb 단백질을 인산화시킵니다. 이 단백질은 다양한 인간 종양에서 과발현되며, 세포 증식 및 분화 조절에 중요한 역할을 합니다. Cyclin D1은 인간 11번 염색체의 CCND1 유전자에 의해 암호화되며, G1 단계에서 합성되어 성장 인자 자극에 따라 CDK4 또는 CDK6과 복합체를 형성합니다. Cyclin D1-CDK 복합체는 Rb 단백질의 성장 억제 기능을 억제하고, CDK 억제제(p21Cip1, p27Kip1)의 활성을 조절합니다. 성장 인자 자극이 없을 경우 Cyclin D1은 불안정해져 유비퀴틴 매개 분해를 통해 G1/S 단계 정지에 관여합니다.
