Product Specific Information
- Heat-mediated antigen retrieval using a sodium citrate buffer (pH 6.0) is recommended for staining of paraffin sections.
- Suggested positive control for immunohistochemical applications: normal lung tissue.
- Mouse anti-human podoplanin antibody, clone 4D5aE5E6, recognizes human podoplanin (also known as gp36, Aggrus, PA2.26 antigen, or T1 alpha).
Target Information
Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type 1 glycoprotein (40–43 kDa) expressed in many tumors and normal cells, especially lymphatic epithelial and follicular dendritic cells. It localizes in stromal cells of peripheral lymphoid tissues and thymic epithelial cells.
As a regulator of lymphatic endothelium, podoplanin likely maintains podocyte shape and serves as a ligand for CLEC-2. Its expression correlates with higher tumor invasiveness and metastatic potential. Podoplanin contributes to cell migration, metastasis formation, and tumor invasion.
Expression is induced through Src activation of Cas and has been observed on Th17 cells. Studies show podoplanin presence in a subset of CD4+ cells from human PBMCs. Dysfunction of podoplanin is linked to diseases such as lymphangioma and subglottic neoplasm. It is also proposed as a marker for lung injury due to its physiological role.
