This antibody reacts with both phosphorylated and non-phosphorylated forms of medium neurofilament protein (160 kDa intracellular antigen) of various species.
Immunocytochemistry protocol:
Carnoy’s fixative 2 × 3 min, blocking with 1% glycine + 0.2% gelatin for 10 min.
Target Information
Neurofilaments (NFs) are intermediate filaments expressed almost exclusively in neuronal cells, predominantly in large axons. They consist of three polypeptide chains — NF-M (medium), NF-H (high), and NF-L (light) — which share structural similarity but differ in terminal sequences.
NF-M’s tail domain mediates interactions critical for axonal size and cytoskeletal architecture, regulated by phosphorylation and O-GlcNAcylation.
Antibodies against neurofilament subunits serve as robust neuronal markers due to their abundance, neuron-specific expression, and biochemical stability.
